An der Universitt von Jena studierte er zunchst von 1754 bis 1758 Theologie, wendete sich jedoch danach der klassischen Philologie zu. 1766 nahm Musus eine literaturkritische Ttigkeit als Mitarbeiter der von Friedrich Nicolai herausgegebenen Allgemeinen Deutschen Bibliothek auf. Dort rezensierte er etwa 350 zeitgenssische Romane nach den Mastben einer aufklrerischen, an der antiken Redekunst orientierten Kunsttheorie. 1769 wurde er zum Professor der klassischen Sprache und Geschichte des dortigen Gymnasiums ernannt. Er war ein Kritiker des Geniekults und der Empfindsamkeit. 1783 wurde er in den Illuminatenorden als sogenannter Presbyter unter dem Namen Dante AlighieriPriscillan aufgenommen. Zu seinen Werken zhlen u. a. : Das Grtnermdchen von Vincennes (1771), Physiognomische Reisen (17781779), Volksmhrchen der Deutschen (5 Bnde) (1782-1786) und Freund Heins Erzhlungen in Holbeins Manier (1785).
